Hepatic enzyme inhibitors of CYP P450 (3A4) like cimetidine.
-
erythromycin, itraconazole, ketoconazole and mibefradil tend to
reduce metabolism and clearance of Sildenafil, this leads to an
increase in Sildenafil plasma level, so use of 25 mg dose is
recommended for patients taking these medications.

Hepatic enzyme inducers of CYP P450 like rifampin, they
decrease Sildenafil plasma concentration since they increase the
metabolism and clearance of Sildenafil.

Concomitant use of nitrates and Sildenafil can lead to an
increase in hypotensive effect.

Single doses of antacids did not affect the bioavailability of
sildenafil.
|
 |
Pharmacokinetic data from patients in clinical trials showed no
effect on sildenafil pharmaco kinetics of:

CYP 2C9 inhibitors ( Such as tolbutamide, warfarin)..
-
CYP2D6 inhibitors (SSRIs, TCAs)
-
Thiazide and related diuretics.
-
ACE Inhibitors and calcium channel blockers.

Sildenafil (5Omg) did not potentiate the increase in bleeding
time caused by aspirin (150mg).

Siidenafil (50mg) did not potentiate the hypotensive effect of
alcohol in healthy volunteers with mean maximum blood alcohol
levels of 0.08%.
